Abstract

Toxoplasma gondii infection is a parasite zoonosis with worldwide distribution which public health impact is related to congenital, ocular and cerebral toxoplasmosis.To date, a variety of serological diagnostic techniques had been implemented and adapted to study this parasitosis; however, few studies about their evaluation and validation have been carried.The aim of this research was the standardization and evaluation of an in house ELISA for IgG antibodies against T.gondii in Peruvian population.Was evaluated a sample of 178 Peruvian sera, which were cataloged as positives or negatives in comparison to a ELISA reference test.To determine cut-off and diagnostic parameters of the in house ELISA we employed ROC curves.Also we established inter and intra assay variation coefficients.The result was a cut-off was a reactivity index of 1.0; with 85.9% sensibility, 70.1% specificity, 65.6% positive predictive value and 88.2% negative predictive value.A correlation inter and intra assay correlation of 0.93, 0.88, 0.57, 0.95, and 0.975, respectively, was obtained.The in house ELISA evaluated and standardized for IgG antibodies against T.gondii in Peruvian population, is a technique with high sensitivity, thus can be used in screening or seroprevalence studies; besides it is easy to perform and of low cost.
